Active pharmaceutical ingredients (APIs) with elongated, needle-shaped crystal structures pose significant manufacturing challenges, often causing poor powder flow, non-uniform die filling, and tablet defects such as capping and lamination due to elastic recovery during decompression. While micronization can reduce particle size, it frequently fails to resolve the underlying mechanical and deformation constraints of high-aspect-ratio crystals. Transitioning from traditional high-shear wet granulation to a Quality by Design (QbD)-guided dry granulation process using roller compaction effectively restructures particle morphology into dense, irregular agglomerates.

This mechanical transformation enhances bulk density and flowability, enabling formulations to achieve target tensile strengths exceeding 2.0 MPa under standard compression forces. Explore the full case study to learn how early morphological characterization and strategic process selection optimize solid dosage development.
