When High-Stakes ADC Decisions Can't Wait for More Material or Better Data

Early-stage antibody-drug conjugate (ADC) development often requires critical decisions to be made with limited material and incomplete analytical data. Among the biggest challenges are obtaining accurate concentration and drug-to-antibody ratio (DAR) measurements, particularly when novel linker-payload chemistries alter traditional UV-based calculations. Conventional approaches frequently rely on separate calibration assays or resource-intensive analytical methods that can slow candidate evaluation and consume valuable samples.

A streamlined workflow offers an alternative by combining LC-MRM with a single UV280 measurement to determine linker-payload extinction coefficients while simultaneously establishing ADC concentration. Validated against an orthogonal UV/Vis method, the approach delivers accurate results using minimal material and eliminates the need for separate calibration assays.
