The Art Of Science And Topical Formulations

Source: MedPharm

In this comprehensive talk, John Lin, Chief Scientific Officer at MedPharm, explores the intricate balance between artistry and science in topical skin formulations. He emphasizes that for a topical product to succeed, especially one applied daily, the formulation must feel cosmetically pleasant to users while also delivering active compounds effectively. This dual requirement makes the formulator’s role uniquely challenging—akin to being both a chef and a scientist.

Lin explains that skin, while protective, is semi-permeable, making it a viable route for drug delivery—if the formulation is right. MedPharm utilizes a range of innovative preclinical models to assess and optimize formulations before clinical trials. These include in vitro release testing to understand compound release kinetics, in vitro permeation testing (IVPT) using human surgical skin to assess delivery into and through tissue, and ex vivo skin culture that keeps tissue viable for up to 30 days—enabling studies on irritation, efficacy, inflammation, and even wound healing.

Their proprietary techniques allow precise control, such as maintaining skin barrier integrity, quantifying drug delivery into specific layers, and simulating complex disease states like psoriasis. They also assess compound activity using molecular biology tools (PCR, ELISA, RNA-seq) to provide a full pharmacokinetic-pharmacodynamic (PK-PD) picture.

Lin also highlights MedPharm’s broader capabilities across other challenging delivery routes, such as nail, ocular, and intranasal systems. With over 25 years of experience, MedPharm positions itself as a full-service CDMO that can support clients from early formulation through to commercial manufacturing, offering unmatched expertise in niche drug delivery.
