Pure Synthetic oDNA For mRNA Therapies: Faster, Higher-Quality, More Efficient mRNA Development

Plasmid linearization and bacterial fermentation are familiar steps in preparing DNA templates for mRNA production. This poster presents a cell-free alternative: synthetic, double-stranded linear DNA that arrives ready for in vitro transcription, with optional adaptors designed around the structure of T7 RNA polymerase to improve its kinetics.
It brings together enzyme kinetics data, pairwise comparisons from more than 30 customer programs, and a case study from a Rift Valley Fever mRNA vaccine project run by Afrigen and funded by CEPI. The authors, from Syngoi, Afrigen, DNA Script, Ribbon, and CIMA at the University of Navarra, assess yield, integrity, dsRNA, and functional response against linear plasmid DNA.
