A suite of engineered circular DNA formats is presented, each designed to improve gene‑editing stability, efficiency, and tolerability. One architecture features a short stabilizing stem that supports strong homology‑directed repair while maintaining high cell viability. Additional options include fully single‑stranded, hybrid, and fully double‑stranded circular configurations, all customizable and capable of carrying complex or large genetic payloads. The reduced immunogenicity of these formats enhances suitability for in vivo applications, especially when paired with lipid‑based delivery systems where double‑stranded DNA often triggers immune pathways.

High knock‑in rates have been demonstrated across multiple primary immune‑cell donors, indicating consistency and reliability. These formats are compatible with a range of delivery approaches such as electroporation, nucleofection, and lipid nanoparticles.
