Improving Accuracy In Size Measurements Of Nanomedicine
By Dr. Maria Marioli, Principal Scientist & Group Lead, Nanomedicine | Ardena

Accurate nanoparticle sizing dictates the success of advanced drug product development. Conventional dynamic light scattering (DLS) often overestimates particle size or masks distinct size populations due to resolution limits. Coupling asymmetric flow field-flow fractionation with phase and spatially-resolved dynamic light scattering (AF4-PhaSR-DLS) resolves these critical analytical bottlenecks.
By separating sample populations prior to detection, this advanced method eliminates common flow-induced artifacts and measurement inconsistencies caused by low sample concentrations—issues that frequently compromise conventional online DLS for particles exceeding 100 nm. Precise characterization prevents misleading conclusions regarding drug bioavailability, filterability during manufacturing, batch-to-batch consistency, and overall product stability. Addressing these analytical limitations de-risks development and ensures robust quality control from early research through full-scale manufacturing.
