With only 6.7% of Phase 1 drugs reaching approval, according to Norstella data, the margin for strategic missteps in drug development is essentially zero. A Drug Development Plan (DDP), sometimes called an Integrated Development Plan (IDP), is the cross-functional framework that keeps your program on track from discovery through regulatory submission and commercial launch.

What you might not realize: a DDP isn't a document you compile from existing data. It works in reverse. You start with the go/no-go decision criteria your Phase 1 must satisfy, then build every protocol, CMC deliverable, and biomarker endpoint backward from those thresholds. Teams that build forward from available data aren't writing a plan; they're writing a summary.

This guide walks through what a DDP actually requires: 40 categorized source documents across strategy, nonclinical, clinical, CMC, regulatory, and commercial domains, 15 of which are IND/CTA-blocking and non-negotiable before regulatory submission. It covers who owns each section, how review moves through three tiers from functional leads to the Program Governance Committee, and how change control keeps the living document current without losing audit trail integrity.

It also names the mistakes that quietly kill programs: assumptions buried in narrative prose, CMC timelines disconnected from clinical milestones, regulatory strategy written after the TPP is finalized. Each one is correctable, but only if you know to look for it.
