Comprehensive Characterization And In Vitro Functionality Study Of Small Extracellular Vesicles Isolated By Different Purification Methods From Mesenchymal Stem Cell Cultures
By Davide Zocco, Associate Director of R&D, Cell and Gene Technologies

Two purification routes can start from the same mesenchymal stem cell conditioned medium and still deliver different small extracellular vesicle preparations. This poster from Lonza scientists Marta Venturella, Laura Bianciardi, Ali Navaei, and Davide Zocco sets size-exclusion chromatography against density gradient ultracentrifugation.
The characterization spans nano flow cytometry, Western blotting, TEM, mass spectrometry, and RNA sequencing, covering particle concentration, size, purity, integrity, identity markers, protein cargo, and miRNA profiles. The team then tested the vesicles in HUVEC migration, proliferation, and tube formation assays. Some measures look closely matched between methods, while others separate them, including how much material it takes to produce a measurable response.
Explore the data to see where the two approaches converge, where they part ways, and what that means for in vitro functional readouts.
