Managing Nitrosamine Risk In Pharmaceutical Products: A Practical Guide To Detection, Formation, And Lifecycle Control Strategies

Nitrosamine impurities remain one of the pharmaceutical industry’s most recent quality and regulatory challenges. While initial regulatory attention focused primarily on solvent-related nitrosamines following several high-profile product recalls, today’s expectations extend well beyond residual solvents. Manufacturers must now evaluate and control nitrosamine drug substance-related impurities (NDSRIs). This evaluation includes manufacturing conditions, packaging materials, and even excipient variability. The evaluation is ongoing throughout the drug product lifecycle to demonstrate management of potential nitrosamine formation.
As regulatory guidance continues to evolve, manufacturers are expected to implement product-specific risk assessments, develop highly sensitive analytical methods capable of detecting impurities at extremely low concentrations, and establish scientifically justified control strategies throughout development and manufacturing.

Variability between excipient suppliers and even between lots of the same excipient grade from the same supplier can significantly influence nitrosamine risk, making upstream nitrite monitoring an increasingly valuable component of an overall control strategy.
